Pharmacokinetics:
Oral absorption is rapid, but not complete. Oral absorption is 50%, reaching the peak concentration in 2-4 hours. After oral administration, the effect lasts for a long time, up to 24 hours. It is widely distributed in various tissues, and a small amount can pass through the blood cerebrospinal fluid barrier. The distribution volume of healthy people is about 50 ~ 75L. The half-life of the drug is 6-7 hours, and it is mainly excreted from the urine in the original form. When the renal function is damaged, the half-life of the drug is prolonged and can be accumulated in the body. It can be removed during hemodialysis. It has low fat solubility, low permeability to brain tissue, and low plasma protein binding rate (6 ~ 16%).
Pharmacology and Toxicology:
It is a selective β 1-adrenoceptor blocker and has no membrane stabilization and endogenous sympathomimetic activity. But it did not inhibit the bronchiectasis of isoproterenol. The mechanism of lowering blood pressure and reducing myocardial oxygen consumption is the same as that of propranolol. Large scale clinical trials have confirmed that atenolol can reduce the mortality of acute myocardial infarction from 0 to 7 days. The therapeutic dose had no significant effect on myocardial contractility.
Matters needing attention:
The clinical effect of this product may not be completely parallel to the blood concentration, and the dose adjustment shall be subject to the clinical effect; the dose shall be reduced when the renal function is damaged; when patients with heart failure symptoms use this product, they shall be given digitalis or diuretics; if the symptoms of heart failure still exist, they shall be gradually reduced; the withdrawal process of this product shall be at least 3 days, and it can often reach 2 days If there are withdrawal symptoms, such as angina pectoris attack, the drug should be given again temporarily and stopped gradually after it is stable; it does not affect its bioavailability when combined with diet; this product can change the tachycardia caused by the decrease of blood glucose; hypertensive patients with chronic obstructive pulmonary disease should use it with caution; this drug can cause peripheral arterial blood circulation disorder, and the patients may be given the conventional dosage for the treatment of allergic reaction There was no response to epinephrine.
